How do I know how many subscribers I have to my podcast? Am I just looking for the number of Downloads that I have from my mp3 files on my sites stats page? Of course this would help me get more advertisers. I do have one and have had as many as three on one show.

Well, if your using FeedBurner your looking at the Feed Circulation.

The circulation on feedburner also logs aggrigator hits to the rss and isn't totally accurate.

I've always seen it as how many times a person walk by the window is feedburner stats. How many times someone purchases the dress in the window is a download.
I internvewed feedburner at the expo, and they put a different slant on it

I used to think feedburner worked that way untill I head something Rob Walch say (not sure if it was on his podcast or another) and it seemed to clear things up quite a bit. Nice interview and cool show. I am now a subscriber!
